运动性心脏肥大发生过程中心房肌内皮素基因的表达

(1.洛阳师范学院 体育学院,河南 洛阳471022; 2.国家体育总局 体育科学研究所,北京100061;3.北京体育大学 运动人体科学学院,北京100084) 摘要:为进一步探讨运动性心脏肥大发生的机制,采用反转录聚合酶链式反应(RT-PCR)技术,对70只SD大鼠进行75 d耐力训练过程中内皮素信使核糖核酸( ET-1mRNA)的表达量进行观察。以异硫氰酸胍法提取心肌组织总核糖核酸(total RNA),测定心房肌中ET-1mRNA,以肌动蛋白(β-actin)为内参照,测定ET-1mRNA与β-actin mRNA的比值为每个测试样中ET-1mRNA表达量。结果显示:在5个时相点中,实验组心房肌组织ET-1mRNA的表达量与对照组相比差异没有显著性意义。在中等强度运动造成的运动性心脏肥大发生过程中,心房肌组织的ET-1mRNA的表达量可能不发生变化。 关键词:内皮素; 心房肌组织; 内皮素信使核糖核酸; 运动性心脏肥大; 大鼠 中图分类号:G804.7 文献标识码:A 文章编号:1006-7116(2006)03-0047-04

(1.College of Physical Education, Luoyang Teacher Institue, Luoyang 471022,China;2.Institute of Sport Science, General Administration of Sport,Beijing 100061,China;3.College of Sport Science of Human Body, Beijing Sport University,Beijing 100084,China) Abstract: In order to probe further into the mechanism of the occurrence of kinetic cardiac hypertrophy, the authors observed the expression of edotheline messenger ribonucleic acid (ET-1 mRNA) of 70 SD rats during their 75 d endurance training by applying reverse transcription polymerase chain reaction(RT-PCR)technology, applied isosulfocyanic acid guanidine approach to abstract total RNA in cardiac muscle tissue and measure ET-1 mRNA in atria muscle, based the internal control on β-actin to measure the ratio of ET-1 mRNA/β-actin mRNA as the express of ET-1 mRNA in each test sample, and revealed that in the 5 time phase points the difference between the expression of ET-1 mRNA in atria muscle tissue of rats in the experimental group and that of rats in the control group had no meaning of significance. During the occurrence of kinetic cardiac hypertrophy caused by medium intensity exercises, the expression of ET-1 mRNA in atria muscle tissue may not be changed.
